Because of the one-way conversions of Third Avenue, north of 24th Street, to northbound, and Lexington Avenue to southbound, several bus route changes take effect:
- The Fifth Avenue Coach Lines - NYCO Division routes 3 (Lexington Avenue) and 4 (Lexington and Lenox Avenues) are discontinued.
- Surface Transit's M-101 route now travels north along Third Avenue below 125th Street, and south on Lexington Avenue between 125th Street and 23rd Street. (Also, a new short-run variant of M-101, running between 23rd and 125th Streets and driven by Fifth Avenue Coach drivers, basically a replacement for the 3 line, is inaugurated.)
- Fifth Avenue Coach Lines - NYCO Division's 2 [Fourth and Madison Avenues (via Madison Avenue)] line is extended from its prior terminus of 135th Street and Madison Avenue, to 146th Street and Lenox Avenue via 116th Street, to make up for the loss of FACL-NYCO Division's 4. (The 1 [Fourth and Madison Avenues (via Park Avenue)] route still terminates at 135th and Madison at this point; it is not until March 1, 1964 that its north end is extended to 146th and Lenox via 135th Street.)
[The 101A (Third, Lexington and Lenox Avenues) route would not come into being until March 2, 1969, folks.]

In addition, Fifth Avenue Coach Lines - FACO Division's 15 - Jackson Heights line, on northbound/eastbound runs, is rerouted north on Third Avenue between 57th and 59th Streets (it had previously run north on Lexington in that stretch).
